چکیده

RSK [for 90 kDa ribosomal S6 kinase (p90rsk); also known as MAPKAP-K1] and MSK (for mitogenand stressactivated protein kinase) constitute a family of protein kinases that mediate signal transduction downstream of MAP kinase cascades. RSK is activated by MAP kinases of the extracellular signalregulated kinase (ERK) family in response to growth factors, many polypeptide hormones, neurotransmitters, chemokines and other stimuli. MSK is also activated by ERK in response to such stimuli, but in addition, it can be activated by p38 family MAP kinases in response to various cellular stress stimuli and proinflammatory cytokines/factors. In mammals, four RSK genes (RSK1-RSK4) and two MSK genes (MSK1 and MSK2) have been identified. RSK and MSK are ubiquitously expressed and many cell types express several members of each family. At present, little is known about specific and overlapping functions of the individual RSK/MSK family members. Orthologues of RSK and MSK have been described in D. melanogaster and C. elegans, but not in yeast or plants.

Contribution of S6K1/MAPK Signaling Pathways in the Response to Oxidative Stress: Activation of RSK and MSK by Hydrogen Peroxide

Cells respond to different kind of stress through the coordinated activation of signaling pathways such as MAPK or p53. To find which molecular mechanisms are involved, we need to understand their cell adaptation. The ribosomal protein, S6 kinase 1 (S6K1), is a common downstream target of signaling by hormonal or nutritional stress. Here, we investigated the initial contribution of S6K1/MAPK si...

Nur77 is phosphorylated in cells by RSK in response to mitogenic stimulation.

Nur77 is a nuclear orphan receptor that is able to activate transcription independently of exogenous ligand, and has also been shown to promote apoptosis on its localization to mitochondria. Phosphorylation of Nur77 on Ser354 has been suggested to reduce ability of Nur77 to bind DNA; however, the kinase responsible for this phosphorylation in cells has not been clearly established. In the prese...

BI-D1870 is a specific inhibitor of the p90 RSK (ribosomal S6 kinase) isoforms in vitro and in vivo.

Hormones and growth factors induce the activation of a number of protein kinases that belong to the AGC subfamily, including isoforms of PKA, protein kinase B (also known as Akt), PKC, S6K p70 (ribosomal S6 kinase), RSK (p90 ribosomal S6 kinase) and MSK (mitogen- and stress-activated protein kinase), which then mediate many of the physiological processes that are regulated by these extracellula...

I-7: Maternal Signalling to the Placenta

Background: Though it is well established that maternal blood-borne signals influence highly the growth of the placenta, the mechanisms are not known. In vitro trophoblast culture models are limited by an inability to reconstruct the polarised bilayer of the human hemochorial placenta. We have used a first trimester villous tissue explant system to investigate how growth factors interact with p...

Human melanoma cells resistant to MAPK inhibitors can be effectively targeted by inhibition of the p90 ribosomal S6 kinase

The clinical availability of small molecule inhibitors specifically targeting mutated BRAF marked a significant breakthrough in melanoma therapy. Despite a dramatic anti-tumour activity and improved patient survival, rapidly emerging resistance, however, greatly limits the clinical benefit. The majority of the already described resistance mechanisms involve a reactivation of the MAPK signalling...
